Im Auftrag von Weyland Industries, vertreten durch Meredith Vickers, macht sich eine Forschergruppe und der Androide David auf eine Reise nach den Ursprüngen menschlichen Lebens.

Angeführt wird die Gruppe von den Forschern Elizabeth Shaw und Charlie Holloway, die davon überzeugt sind, dass die mysteriösen Zeichen, die sie weltweit entdeckt haben, sie zu wertvollen Antworten über den Beginn der Menschheit führen.

Die zweijährige Expedition führt die Passagiere der "Prometheus" schließlich zu den dunkelsten Ecken des Universums, wo sie Schlimmes erwartet...
